Proteasome selectivity towards Michael acceptor containing oligopeptide-based inhibitors.
The synthesis and biological evaluation of ten Michael acceptors containing potential proteasome inhibitors are described. Cellular targets of azide containing inhibitors and were assessed in HEK293T and RAW264.7 cells by a two step labeling strategy, followed by biotin-pulldown, affinity purification, on-bead tryptic digestion and LC-MS(2) identification. This strategy appears to be an attract...
متن کاملNew Bioactive Diterpenoid from Euphorbia decipiens
One new diterpene ester with a tricyclic lathyrane-or myrsinol-type skeleton has been isolated fromEuphorbia decipiensBoiss. &Buhse. In addition to one new, the known constituents -amyrin, -amyrin acetate, methyl (2,4-dihydroxy-3-formyl-6-methoxy) phenyl ketone (2), and 1,1-bis (2,6-dihydroxy-3-acetyl-4-methoxyphenyl)methane (3) have been isolated from the same source. The structure elucidation...
متن کاملA triphenylamine-containing donor-acceptor molecule for stable, reversible, ultrahigh density data storage.
In the past few years, there has been increasing interest in the application of organic functional molecules for next generation information storage and future optoelectronic devices.1 With the development of the scanning probe microscopy (SPM), ultrahigh density data storage based on SPM technology has made great progress in recent years.2 Among these studies, organic functional materials have...
متن کاملModel studies toward the synthesis of the bioactive diterpenoid, harringtonolide.
In model studies towards the synthesis of harringtonolide, the construction of the tropone moiety via arene cyclopropanation was investigated. The installation of the lactone ring was accomplished by way of a Diels-Alder cycloaddition of various indenones and a-pyones. The incorporation of the key bridge methyl group and subsequent control of its stereochemistry is also outlined.
متن کاملRegulation of human dihydrodiol dehydrogenase by Michael acceptor xenobiotics.
A human oxidoreductase (H-37) that is overexpressed in ethacrynic acid-resistant HT29 colon cells (Ciaccio, P. J., Stuart, J.E., and Tew, K.D. (1993) Mol. Pharmacol. 43, 845-853) has been identified as a dihydrodiol dehydrogenase. Translated protein from a dihydrodiol dehydrogenase cDNA isolated from a library prepared from ethacrynic acid-resistant HT29 cell poly(A+) RNA was recognized by anti...
